Title: Mass Spectrometry Analysis of Mucin O-Glycosylation in Alzheimer's Disease
Summary: My research focuses on the optimization of mass spectrometry-based workflows for the study of mucin-type O-glycosylation in Alzheimer's disease. First, I characterized the mucinase SmE for use in MS. SmE is a remarkably powerful enzyme that enabled in-depth glycoproteomic analysis of a suite of biologically-relevant proteins, outperforming commercially available options. Next, I applied this enzyme to perform O-glycoproteomic analysis of aged rhesus macaques, a uniquely relevant model for sAD. Finally, I took a more focused approach by studying the O-glycosylation on amyloid precursor protein (APP) and investigating the effects of this modification on pathogenic processing.
